This investigation utilized a mouse model with diminished social interactions, providing critical insights into the substance's mechanisms.
Published in Nature Communications, the study examined the compound's capacity to boost social engagement in mice genetically predisposed to reduced sociability, specifically a Cntnap2-knockout model. Intriguingly, the research indicated that psilocybin enhanced social behaviors in these mice but not in those exhibiting normal social interaction levels. This suggests a dependency on the brain's initial behavioral state for the psychedelic's lasting influence.
The researchers also tackled a long-standing question: does an individual need to be conscious during the acute psychedelic experience for the therapeutic effects to manifest? To explore this, psilocybin was administered to mice under light anesthesia. The results showed a significant and persistent improvement in sociability, lasting at least two weeks, even without the mice being awake during drug administration. This finding suggests that the therapeutic benefits of psilocybin might not hinge on the subjective conscious experience, which could simplify future clinical applications by potentially mitigating the need to manage acute hallucinogenic states. Furthermore, the study underscored that the enhancement of sociability was mediated by the activation of serotonin 2A receptors, as blocking these receptors prevented the observed effects.
While offering promising insights, the study's authors emphasize the preliminary nature of these findings and caution against direct extrapolation to human therapeutic benefits, particularly for conditions like autism spectrum disorder.
